چکیده

The cytoplasmic pH value o f guard cells o f Valerianella locusta was determined from the distribution o f the weak acid [l4C]ABA between the medium and the cell, which was shown to be 8.1 ±0.1. Treatment o f guard cells in media with low pH (pH 5.0) and different concentra tions o f sodium acetate (0 -5 m o lm '3) caused a considerable acidification o f the cytoplasm (6 .6-7 .7). Using the compartmental analysis we determined also the fluxes o f ABA at the plas ma membrane o f guard cells (co) and at the tonoplast o f guard cells (cv) under steady state conditions. The flux o f ABA from the cytoplasm into the vacuoles (tonoplast flux) was de pendent upon the cytoplasmic pH. The data give evidence that A B A “ is transported across the tonoplast by a saturable transport component. The ABA flux at the plasma membrane seemed to be independent o f the cytoplasmic pH.

Nitric Oxide as a Mediator of Aba Signalling in Stomatal Guard Cells

Water shortage is likely to be one of the major global environmental stresses of the 21st century. A key plant response to declining soil water potential is increased biosynthesis and subsequent action of abscisic acid (ABA). ABA is an endogenous anti-transpirant that induces stomatal closure, thereby leading to water conservation. The signal transduction processes commencing with guard cell AB...

Vacuolar CAX1 and CAX3 influence auxin transport in guard cells via regulation of apoplastic pH.

CATION EXCHANGERs CAX1 and CAX3 are vacuolar ion transporters involved in ion homeostasis in plants. Widely expressed in the plant, they mediate calcium transport from the cytosol to the vacuole lumen using the proton gradient across the tonoplast. Here, we report an unexpected role of CAX1 and CAX3 in regulating apoplastic pH and describe how they contribute to auxin transport using the guard ...

Ca-Dependent and -Independent Abscisic Acid Activation of Plasma Membrane Anion Channels in Guard Cells of Nicotiana tabacum

Drought induces stomatal closure, a response that is associated with the activation of plasma membrane anion channels in guard cells, by the phytohormone abscisic acid (ABA). In several species, this response is associated with changes in the cytoplasmic free Ca concentration. In Vicia faba, however, guard cell anion channels activate in a Ca-independent manner. Because of potential differences...

Are diurnal patterns of stomatal movement the result of alternating metabolism of endogenous guard cell ABA and accumulation of ABA delivered to the apoplast around guard cells by transpiration?

Abscisic acid (ABA) prevents opening of closed stomata and causes open stomata to close. A dual-source model is proposed linking ABA to diurnal stomatal movements. Darkness would favour guard cell biosynthesis of endogenous ABA and disfavour ABA catabolism. At first light, xanthophyll cycling, isomerization of ABA precursors, and activation of a cytochrome P450 mono-oxygenase (CytP450) would de...

Nitric oxide in guard cells as an important secondary messenger during stomatal closure

The modulation of guard cell function is the basis of stomatal closure, essential for optimizing water use and CO2 uptake by leaves. Nitric oxide (NO) in guard cells plays a very important role as a secondary messenger during stomatal closure induced by effectors, including hormones. For example, exposure to abscisic acid (ABA) triggers a marked increase in NO of guard cells, well before stomat...
